Design Notes

Designed based on the scar sequences formed between RFC10 parts when joined, to preserve the RBS-start codon spacing and sequence context and the sequence context upstream of the RBS, which affects translation initiation rate. The -1 nucleotide to the start codon is A to form the AATG "scar" that should overlap with the start codon.
